<p>Stoke Therapeutics (Nasdaq: STOK) is a biotechnology company dedicated to restoring protein expression by harnessing the body’s potential with RNA medicine. <p>STK-002 is Stoke’s proprietary antisense oligonucleotide (ASO) in clinical development for the treatment of autosomal dominant optic atrophy (ADOA), the most common inherited optic nerve disorder. STK-002 has been granted orphan drug designation by the FDA. A Phase 1 study (OSPREY) of STK-002 in people with ADOA is now underway. The company is also conducting early research in Syngap1, a severe and rare neurodevelopmental disorder, in collaboration with Acadia Pharmaceuticals.</p>
<p>Stoke’s initial focus is on diseases of the central nervous system and the eye that are caused by a loss of ~50% of normal protein levels (haploinsufficiency). Proof of concept has been demonstrated in other organs, tissues, and systems, supporting broad potential for the Company’s proprietary approach.</p></div><p><strong><u>Position Purpose:</u></strong></p>
<p>The Senior Scientist will work collaboratively across the Research organization to support preclinical discovery efforts to understand the pharmacokinetics, pharmacodynamics, and efficacy of novel compounds in disease models. This position requires a scientist with deep expertise designing and developing robust assays to investigate preclinical pharmacokinetic, pharmacological, and functional biomarkers in tissue samples. This position will report into the Executive Director of in vivo pharmacology.</p>
<p><strong><u>Key Responsibilities:</u></strong></p>
<ul>
<li>Develop and optimize preclinical PK,PD, and biomarker assays to support preclinical research efforts and bridge into early development activities.</li>
<li>Use a high level of scientific judgment and creative thinking to adapt and/or create standard methods by applying breadth of knowledge and prior work experience.</li>
<li>Work collaboratively within and across functions to support programs from early discovery through preclinical proof of concept and candidate selection to advance the company pipeline.</li>
<li>Maintain excellent and timely recordkeeping while adhering to aggressive timelines</li>
<li>Author study reports that may be used for regulatory filings and scientific manuscripts.</li>
</ul>
<p><strong><u>Required Skills &amp; Experience: </u></strong></p>
<ul>
<li>Bachelor’s degree with 16+ years of experience, or Master’s degree with 10+ years of post-graduate experience, or Ph.D<strong><em>.</em></strong> with 6+ (industry experience is required). Degrees in Pharmacology, Neuroscience or relevant disciplines preferred.</li>
<li>Deep experience developing and optimizing assays (i.e MSD, ELISA, H-ECL, LCMS, qPCR) in complex matrices, IHC skills a plus.</li>
<li>Experience working in drug discovery with an emphasis on neurology indications, experience working with oligonucleotides preferred.</li>
<li>Designing and managing experiments at contract research organizations.</li>
<li>Experience with PK/PD modeling a plus.</li>
<li>Excellent verbal and written communication skills.</li>
<li>Excellent planning, organization and time management skills including the ability to support and prioritize multiple projects.</li>
</ul>
<p><strong><u>Location(s): </u></strong></p>
<p>Stoke is located in Bedford, MA, and will be moving to a new location in Waltham, MA by the end of 2026. This position is lab-based currently based in Bedford and will later be based in Waltham.</p>
<p><strong><u>Travel:</u></strong></p>
<p>This position will require approximately 5% travel.</p>

<p>The anticipated salary range for this role is $155,00 - $179,000.<strong> </strong>The final offer will be determined based on a variety of factors, including role scope, experience and qualifications, education, market benchmarks, internal equity, and geographic location. In addition to base salary, Stoke offers an annual bonus and equity participation.</p>
